Abstract:
Mass spectrometry (MS) is an indispensable technique in proteomics research. We have pioneered an advanced method, nDIA, which synergistically integrates a Quadrupole Orbitrap mass spectrometer with the Astral analyzer. This innovation facilitates swift and precise protein scanning. Recognizing the intricacies and potential pitfalls inherent to conventional MS methodologies, we have conceptualized and executedOne-Tip, a streamlined workflow. By employing a mere two-step protocol with an Evotip, sample preparation is expedited to an hour. This methodology not only mitigates errors but is also efficient with lower cell input and is adept at processing a vast number of samples daily. With a sample of approximately 1,000 cells, it identified more than 9,000 proteins, mirroring the accuracy of traditional methods. Further, its application on mouse embryos yielded superior protein detection rates, enriching our understanding of embryonic development. A subsequent application on individual HeLa cells led to the identification of over 5,000 proteins, positioning it as the premier methodology for single-cell proteomics.
