[Lecture] Going to the heart of heart muscle

Abstract: 

Cardiac excitation-contraction coupling requires dads, the nanoscopic micro-domains formed adjacent to Z-lines by apposition of transverse tubules and junctional sarcoplasmic reticulum. Dyads are frequently disrupted in heart failure, and lack of dyads is a central limitation of cardiomyocytes derived from human induced pluripotent stem cells (hiP-SC-CMs). However, little is known about the mechanisms that modulate dad organization during cardiac development, homeostasis, and disease. We developed proximity proteomics in intact, living hearts to identify proteins enriched near dyads. Using genetically modified mouse and hiPSC-CMs models, we identified a signaling pyramid that establishes and maintains the architecture of dyads, based on which we developed a new gene therapy approach for heart failure, as well as a methodology for in vitro hiPSC-CMs maturation. This presentation will discuss how master proteins regulate dad structure and function using CMYA5 as the main example.
